GCMS is a technique to separate and analyse compounds that are non-polar and volatile. Examples include hydrocarbons, plant metabolites from essential oil. However, fatty acids, polar organic acids, plant secondary metabolites need to be derivatised before subjecting it to GCMS analysis. Derivatisation reduces polarity and improves stability of the compounds. Identification of the compounds is based on the mass spectral match between experimental data with that of the mass spectra in GCMS library (NIST). GCFID is used as a quantitative tool to quantify compounds based on the retention time (RT) of respective standards.

HPLC is an analytical technique to separate polar compounds in order to purify them from crude extracts, study enzyme kinetics using UV as the detector. A variety of column chemistries are available in the market for various applications.

 

                                                           

 

Services rendered by the facility:

  1. Sample analysis using the above mentioned instruments are provided to students/post docs from the various departments of IISc.
  2. Sample analysis using GCMS is also provided to academic institutions across India
  3. Data analysis using vendor specific softwares and Database (AMDIS, NIST, Data Analysis software, EZ Chromelite)
  4. Sample preparation  including derivatisation of polar compounds for GCMS
  5. Guidance to students and other researchers on various research projects

  • Method optimization to separate D and L amino acids to check activity of racemase from bacteria
  • Method optimization to separate oligonucleotides extracted from bacteria
  • Identification and quantitation of secondary metabolites (carotenoids, taxols, alkaloids, diterpenoids) with reference to standards
  • Enzymatic assay using atropine as the substrate
  • Checking the purity of a protein
  • Protease activity of a viral protein
  • Enzymatic assay using NDP bound sugar as the substrate
  • Separation of proteins using size exclusion chromatography at various pH
